Sıçan Ovaryum Dokusunda Doksorubisin ile İndüklenmiş Folliküler Apoptotik Aktivasyonda Dialil Disülfit’in Koruyucu Etkisi

Öz

Bu çalışmada önemli bir kemoterapötik ajan olan doksorubisinin ovaryum üzerindeki toksisitesini belirlemek ve ovaryan toksisite üzerine yüksek antioksidan kapasiteye sahip olan dialil disülfitin etkinliğini değerlendirmek amaçlandı. Çalışma kapsamında 35 adet Wistar albino cinsi dişi sıçan 5 alt gruba ayrıldı: Grup I: Kontrol-Doksorubisin Grubu, Grup II: Kontrol-Dialil disülfit Grubu, Grup III: Doksorubisin Uygulanan Grup, Grup IV: Dialil disülfit Uygulanan Grup, Grup V: Doksorubisin + Dialil disülfit Uygulanan Grup. Morfolojik değerlendir-me hematoksilen eozin boyaması ile gerçekleştirildi. Apoptotik aktivasyon TUNEL yöntemi ile değerlendirildi. Doksorubisin uygulamasının primordiyal, sekonder ve graaf follikül sayılarında azalmaya sebep olduğu gözlemlenirken, atretik follikül sayılarını ise arttırdığı saptandı. Dialil disülfit tedavisinin doksorubisin ile indüklenmiş ovaryan toksisiteyi azaltabileceği ve ovaryan follikül rezervinin korunmasında etkili olabileceği sonucuna varıldı.

Effect of Diallyl Disulfide on Doxorubicin-induced Follicular Apoptotic Activation in Rat Ovarian Tissue

Öz

The aim of this study was to determine the toxicity of doxorubicin, an important chemotherapeutic agent, on the ovary and to evaluate the efficacy of dialyl disulfide which has high antioxidant capacity on ovarian toxicity. In this study, 35 female Wistar albino rats were divided into 5 subgroups: Group I: Control-Doxorubicin Group, Group II: Control-Dialyl disulfide Group, Group III: Doxorubicin Applied Group, Group IV: Diallyl disulfide Applied Group, Group V: Doxorubicin + Diallyl disulfide Group. Morphological evaluation was performed with hematoxylin eosin staining. Apoptotic activation was evaluated by TUNEL method. Doxorubicin caused a decrease in the number of primor-dial, secondary and graaf follicles, and increased atretic follicle counts. It was concluded that dialyl disulfide treatment may reduce doxoru-bicin-induced ovarian toxicity and may be effective in maintaining ovarian follicular reserve.
