Scilla siberica subsp. armena Yaprak Sapından İn Vitro Çoklu Sürgün Rejenerasyonu

Abstract

Scilla siberica subsp. armena çok yıllık, mavi çiçekli soğanlı bir bitkidir. Bu çalışmada Scilla siberica subsp. armena’nın yaprak sapı kullanılarak; 0.25, 0.50, 1, 2 mg/L TDZ ve 0.1, 0.2 mg/L NAA içeren (8 kombinasyon) MS besin ortamında etkili bir çoklu sürgün rejenerasyon sistemi geliştirmek amaçlanmıştır. Maksimum ortalama rejenere sürgün yüzdesi 2 mg/L TDZ + 0.1 mg/L NAA içeren MS besin ortamında kaydedilmiştir. Maksimum eksplant başına düşen ortalama sürgün sayısı 2 mg/L TDZ + 0.2 mg/L NAA içeren MS besin ortamında raporlanmıştır. Maksimum ortalama sürgün uzunluğu 0.25 mg/L TDZ + 0.2 mg/L NAA ilave edilen MS besin ortamından elde edilmiştir. Gelişen sürgünler 1 mg/L IBA içeren MS besin ortamında köklendirilmiştir.

In Vitro Multiple Shoot Regeneration from Scilla siberica subsp. armena Petiole

Abstract

Scilla siberica subsp. armena is a perennial blue flowered bulbous plant. This study aimed to develop an efficient multiple shoot regeneration system for S. siberica subsp. armena using petiole on MS medium containing 0.25, 0.50, 1, 2 mg L-1 TDZ and 0.1, 0.2 mg L-1 NAA (8 combinations). Maximum mean shoot regeneration percentage was noted in MS medium containing 2 mg L-1 TDZ + 0.1 mg L-1 NAA, maximum mean number of shoot per explant was recorded in MS medium containing 2 mg L-1 TDZ + 0.2 mg L-1 NAA, maximum mean number of shoot length was recorded in MS medium supplemented with 0.25 mg L-1 TDZ + 0.2 mg L-1 NAA. Well developed micro propagated shoots were rooted on MS medium containing 1 mg L-1 IBA.
