Pırlak Koyunlarında BMPR1B, BMP15 ve GDF9 Genlerinde Olası Polimorfizmlerin Araştırılması

Abstract

Bu
araştırmada, Afyonkarahisar İli Damızlık Koyun-Keçi Yetiştiricileri Birliği
damızlık istasyonunda 2016 ve 2017 yıllarında çoklu
ve tekli doğum yapanlardan sekizer baş olmak üzere
toplam 16 baş Pırlak koyunda Bone Morphogenetic Protein Receptor type 1B
(BMPR1B), Bone Morphogenetic Protein 15 (BMP15) ve Growth Differentiation
Factor 9 (GDF9) genlerindeki polimorfizmin varlığı ve bunun tespit edilmesi
durumunda çoklu doğumla ilişkilendirilmesi amaçlanmıştır. Genetik
çalışmalarda kullanılmak üzere toplanan kan örnekleri hayvanların V. jugularis’lerinden EDTA’lı vakumlu
tüplere alınmış ve genomik DNA izolasyon kiti kullanılarak DNA izolasyonları
yapılmıştır. İlgili genler yönünden gerçekleştirilen PCR ve DNA dizileme
analizleri sonucunda her üç gen yönünden de Pırlakların monomorfik yapıda
olduğu ortaya konulmuştur.

Investigation of Possible Polymorphisms in BMPR1B, BMP15 and GDF9 Genes in Pırlak Sheep

Abstract

This study was realised to determine the presence of polymorphism in
major proliferacy genes named
Bone Morphogenetic Protein
Receptor type 1B (BMPR1B), Bone Morphogenetic Protein 15 (BMP15) and Growth
Differentiation Factor 9 (GDF9)
of 8 uniparous and 8 multiparous
Pırlak sheep reared at Sheep and Goats Breeders Association’s Breeding Station,
Afyonkarahisar between 2016 and 2017. Blood samples collected for using in
genetic studies were taken from V. jugularis of animals into EDTA vacuum tubes
and DNA isolations were performed using genomic DNA isolation kit. The PCR and
DNA sequencing analyzes carried out on the basis of the respective genes
revealed that Pırlaks are monomorphic in the aspect of above mentioned genes.
