Dianthus carmelitarum Ekstraktının Antioksidan ve Sitotoksik Özelliklerinin İncelenmesi

Abstract

Dianthus carmelitarum Reut. Ex Boiss, Dianthus cinsi ve Caryophyllaceae familyasına mensup bir bitki olup, bu cins bitkiler sıklıkla geleneksel tedavide kullanılmaktadırlar. Farklı Dianthus türlerinin farklı ekstraktlarının antioksidan ve sitotoksik etkilerini konu alan çok sayıda çalışma olmasına rağmen, D. carmelitarum ekstraktlarının bu özellikleri ile ilgili herhangi bir çalışma bulunmamaktadır. Bu çalışmanın amacı D. carmelitarum’un dimetil sülfoksitli ekstraktının antioksidan özelliklerini ve sitotoksik etkilerini belirlemektir. Ekstraktın toplam fenolik madde miktarı, toplam flavonoid madde miktarı ve indirgeyici antioksidan güç tayini spektrofotometrik yöntemler kullanılarak belirlendi. Ekstraktın sitotoksik etkisi ise insan serviks (HeLa), akciğer (A549), meme (MCF-7) ve prostat (PC-3) kanser hücre serilerinde MTT yöntemi kullanılarak değerlendirildi. Ekstraktın toplam fenolik madde içeriği ve indirgeyici antioksidan güç değerleri g örnek başına sırasıyla 853.9±19.4 mg gallik asit eşdeğeri ve 1378.7±87.4 mg troloks eşdeğeri olarak bulundu. Ekstrakt çalışılan tüm kanser hücre serileri üzerinde sitotoksik etki gösterdi ve IC50 değerleri 117.4-142.1 µg/mL arasında hesaplandı. Bu çalışma D. carmelitarum ekstraktının in vitro koşullarda kanser hücre serileri üzerindeki sitotoksik etkisini ortaya koyan ilk çalışmadır. Ekstrakttaki etken molekülleri ve bunların etki mekanizmalarını belirleyebilmek için daha ileri çalışmalara ihtiyaç vardır.
